In vivo Analysis of H2AX Phosphorylation Induced by γ-Radiation
Authors
Abstract:
ABSTRACT Background and Objectives: Exposure to ionizing radiation in modern societies is inevitable and can cause a variety of adverse health effects such as cancer and birth defects. Therefore, a reliable, repeatable and sensitive method is required for evaluation of radiation exposure. The aim of this study was to determine the amount of histone H2AX phosphorylation as an indicator of radiation exposure to evaluate the rate of double-strand DNA breakage in irradiated mice. Methods: In this study, 15 mice were exposed to different doses of ionizing radiation. After extraction of total protein from bone marrow cells, γH2AX protein was measured by western blotting. Data analysis was performed using ANOVA, Tukey's post hoc test, and the Pearson's correlation test. Results: The amount of γH2AX protein in the exposed groups increased significantly compared to the control group (P<0.05). Conclusion: The results of this study indicate that exposure to ionizing radiation increases the amount of γH2AX protein in bone marrow cells during the early hours. The protein can be used as a biomarker for monitoring of acute radiation or suspected local radiation exposure. Keywords: γH2AX Protein, Ionizing Radiation, Mouse.

Expression of phosphorylated histone H2AX in blood lymphocytes of patients undergoing angiographic procedures following exposure to X‐rays
Introduction: Coronary angiography is a Diagnostic-Therapeutic method involving ionizing radiation. This method causes to DNA damage with form double stranded breaks which is followed by the phosphorylation of the histone, H2AX. H2AX is a key factor in the repair process of damaged DNA which will accumulate to damage sites. In human cells, H2AX constitutes about 10% of the H2A ...

15 صفحه اولγ-H2AX as a protein biomarker for radiation exposure response in ductal carcinoma breast tumors: Experimental evidence and literature review
Background: H2AX is a histone variant that is systematically found and ubiquitously distributed throughout the genome. DNA double-strand breaks (DSBs) induce phosphorylation of H2AX at serine 139 (γH2AX), an immunocytochemical assay with antibodies recognizing γH2AX has become the gold standard for the detection of DSBs. The importance of this assay to investigate different individu...
full textRole of á-MSH in Repair of UV-Induced DNA Damage, as Determined by H2AX Phosphorylation
Melanoma is the deadliest form of skin cancer and is on the rise. An important paracrine factor that offers photoprotection against the carcinogenic effect of ultraviolet radiation (UV) is ámelanocortin (á-MSH). It not only stimulates eumelanin synthesis, a photoprotective pigment, but also reduces the generation of damaging reactive oxygen species, and enhances nucleotide excision repair (NER)...
full textIonizing radiation-induced DNA strand breaks and γ-H2AXγ-H2AX foci in cells exposed to nitric oxide.
A number of studies have demonstrated that nitric oxide enhances radiosensitivity of anoxic and hypoxic cells in vitro and in vivo, and some evidence points to a role for DNA damage and repair in this phenomenon. We have recently observed that nitric oxide enhances the formation of DNA single- and double-strand breaks following ionising irradiation, measured by the alkaline comet assay and immu...
